Mechanism of Action
Insulin glargine
Mechanism of action Insulin glargine is a weakly soluble human insulin analogue at neutral pH, totally soluble at acid pH.
After injection into the subcutaneous tissue, the acid solution is neutralized, resulting in the formation of microprecipities from which small amounts of insulin glargine are released continuously.
As a result, the concentration/time curve is regular, without peaks, predictable, and the duration of action is prolonged.
The main effect of insulin, including insulin glargine, is to regulate glucose metabolism.
Insulin and its analogues decrease blood glucose by stimulating peripheral glucose uptake, particularly in skeletal muscles and adipose tissue, and by inhibiting hepatic glucose production.
Insulin inhibits lipolysis in adipocyte, inhibits proteolysis and stimulates protein synthesis.
Clinical pharmacology studies have shown that identical doses of insulin glargine and human insulin, administered by the intravenous route of action, can affect all other insulin parameters.
