Mechanism of action
Rupatadine is a second-generation, long-acting antihistamine with a selective antagonistic effect on peripheral H1 receptors.
Some metabolites (desloratadine and its hydroxylated metabolites) retain antihistamine properties and may contribute partially to the efficacy of the drug.
Inhibition of mast cell degranation following immunological or non-immunological stimuli, as well as inhibition of cytokine release, including TNFalpha in human mastocytes and monocytes, have been observed in in vitro tests with very high levels of rubatadine.
The clinical significance of these experimental observations in vitro remains to be demonstrated.
